This study used HaCaT cells, an immortalised human keratinocyte line, rather than live animals. The cells were stimulated with TNF-α and IFN-γ to mimic the inflammatory environment of atopic dermatitis, then treated with Torilis japonica extract (TJE), GHK-Cu, or a complex of the two. The researchers tested several TJE-to-GHK-Cu ratios and report that a 6:4 ratio produced the greatest reduction in TARC and CTACK chemokine expression and IgE production compared with either component alone. The complex also reduced transcription of IL-4, IL-5, IL-10, and IL-13, showed radical scavenging activity without reducing cell viability, and increased keratinocyte migration in a wound healing assay. The paper reports these as anti-inflammatory, antioxidant, and regenerative effects in this cell model, and calls for further investigation of the formulation for atopic dermatitis.
